The cancer risk associated with the use of antipsychotic medications such as phenothiazines (also called tranquillizers or sedatives) and butyrophenones (also called neuroleptics or antipsychotics) has been recognized for decades. There is consistent evidence in the literature that these medications are associated with a higher than expected risk of diabetes mellitus and hypertension, and a moderately increased risk of cardiovascular disease. Antipsychotics are also associated with an increased risk of bone fractures, stroke, and death. Published epidemiological data indicate that, among persons with schizophrenia, the risk of death from suicide in patients treated with antipsychotic medications is up to four times that of the general population. People with mental illness are also much more likely than the general population to die from infectious diseases, particularly pneumonia and influenza, diseases that are often treated with antipsychotic medications. Finally, the incidence of sudden cardiac death and suicide is twice as high for persons with schizophrenia and other mental illnesses than for the general population. The objective of this study is to examine the risk of cancer associated with the use of antipsychotic medications, especially in relation to the age of the individual at first use and the duration of use. We will also examine the types of cancer most often seen in relation to antipsychotic medication use.
Methods:
A large national cohort of up to 1.5 million patients with schizophrenia or other psychotic conditions will be followed to determine the incidence of cancer among those who were prescribed antipsychotic medications (n=69,000) and those who were not prescribed
