The p21(cip1/waf1) inhibitor of cyclin-dependent kinases is one of the many genes induced by wild-type p53. However, it is unclear whether p21(cip1/waf1) is a significant mediator of p53-dependent apoptosis in ovarian cancer cells resistant to the platinum derivatives cisplatin and carboplatin. Thus, we investigated whether the effect of a cell-permeable inhibitor of p21(cip1/waf1) on cisplatin-sensitive and -resistant cell lines differs. A cell-permeable inhibitor of p21(cip1/waf1) (NSC-23766) synergistically increased apoptosis induced by cisplatin in cisplatin-resistant SKVLB-1 human ovarian cancer cells, compared with p21(cip1/waf1) siRNA treatment. The cisplatin cytotoxic effect was associated with activation of p21(cip1/waf1) and p53, inhibition of CDK2 and CDC2 and apoptosis induction in the absence of DNA fragmentation. SKVLB-1 cisplatin-resistant sublines (SKVLB-1/CisP and SKVLB-1/CP) are resistant to cisplatin-induced apoptosis.
